施氏鲟Asnanos1基因序列分析及其在雌雄不同组织中表达

本研究首先从施氏鲟(Acipenser schrenckii)性腺转录组中获得nanos1(Asnanos1)基因全长c DNA,并对其序列的准确性和特征分别进行PCR验证和生物信息学分析。进一步利用荧光定量RT-PCR技术检测Asnanos1基因在雌、雄施氏鲟的性腺、心脏、鳃、脑、肾、肝脏、脾脏等组织中的表达量。利用荧光定量RT-PCR技术检测Asnanos1基因在雌、雄施氏鲟的性腺、心脏、鳃、脑、肾、肝脏、脾脏等组织中的表达量。结果显示:Asnanos1的c DNA全长为1 389 bp,其中,5′非编码区(UTR)为414 bp,3′UTR为279 bp,ORF为696 bp。该基因共编码231个氨基酸。Asnanos1基因在施氏鲟除心脏以外的各组织中均有表达,在雌、雄鱼的鳃、脑、肾、肝脏、脾脏等组织中表达量无显著差异,但在卵巢中的表达量显著高于其他各组织(P0.05)。所得到的结果可为人工培育全雌施氏鲟苗种提供一些基础资料。  相似文献   

基于规则的位置地图情境推理构建与实例分析

情境在移动位置服务领域起到融合多源时空数据、模拟地图使用情形及获取用户空间需求的作用。根据作者早期建立的位置地图情境模型,本文基于活动理论构建了位置地图情境三层推理规则,重点描述其基本原理与方法;通过形式化表达将情境推理的结果与位置地图表达联系起来,形成三层推理规则的解析与运行流程;以用户去机场登机为例,假设2种不同情形,验证5种情境事例。实验结果表明,本文的情境推理方法简洁、可操作性强,为进一步研究智能化地图服务提供参考和借鉴。  相似文献   

Optimization of the purification methods for recovery of recombinant growth hormone from Paralichthys olivaceus

This study aimed to optimize the purification of recombinant growth hormone from Paralichthys olivaceus. Recombinant flounder growth hormone (r-fGH) was expressed by Escherichia coli in form of inclusion body or as soluble protein under different inducing conditions. The inclusion body was renatured using two recovery methods, i.e., dilution and dialysis. Thereafter, the refolded protein was purified by Glutathione Sepharase 4B affinity chromatography and r-fGH was obtained by cleavage of thrombin. For soluble products, r-fGH was directly purified from the lysates by Glutathione Sepharase 4B affinity chromatography. ELISA-receptor assay demonstrated that despite its low receptor binding activity, the r-fGH purified from refolded inclusion body had a higher yield (2.605 mgL-1) than that from soluble protein (1.964 mgL-1). Of the tested recovery methods, addition of renaturing buffer (pH 8.5) into denatured inclusion body yielded the best recovery rate (17.9%). This work provided an optimized purification method for high recovery of r-fGH, thus contributing to the application of r-fGH to aquaculture.  相似文献

将新加坡石斑鱼虹彩病毒(Singapore grouper iridovirus,SGIV)的ORF162的开放式阅读框插入pET-32a表达载体T7启动子控制下的6-His·Tag编码基因上游,构建SGIVORF162原核表达质粒pET-ORF162。表达质粒转化入大肠杆菌BL21(DE3)菌株,经IPTG诱导,成功表达SGIV ORF162融合蛋白。对IPTG浓度、诱导温度、诱导时间等诱导表达条件进行优化后,确定在0.7mmol/LIPTG、16℃条件下诱导14h时可溶性SGIV ORF162重组蛋白占重组蛋白总量的95%。经镍琼脂糖凝胶纯化,获得纯度为90%以上的SGIV ORF162蛋白。用纯化的SGIV ORF162蛋白免疫小鼠,获得高效特异的SGIV ORF162多克隆抗体。  相似文献

Over the past few decades, earthquake engineering research mainly focused on the effects of strong seismicshaking. After the 1999 earthquakes in Turkey and Taiwan, and thanks to numerous cases where fault rupture causedsubstantial damage to structures, the importance of faulting-induced deformation has re-emerged. This paper, along withits companion (Part Ⅱ), exploits parametric results of finite element analyses and centrifuge model testing in developing afour-step semi-analytical approach for analysis of dip-slip (normal and thrust) fault rupture propagation through sand, itsemergence on the ground surface, and its interaction with raft foundations. The present paper (Part Ⅰ) focuses on the effectsof faulting in the absence of a structure (i.e., in the free-field). The semi-analytical approach comprises two-steps: the firstdeals with the rupture path and the estimation of the location of fault outcropping, and the second with the tectonically-induced displacement profile at the ground surface. In both cases, simple mechanical analogues are used to derive simplifiedsemi-analytical expressions. Centrifuge model test data, in combination with parametric results from nonlinear finite elementanalyses, are utilized for model calibration. The derived semi-analytical expressions are shown to compare reasonably wellwith more rigorous experimental and theoretical data, thus providing a useful tool for a first estimation of near-fault seismichazard.  相似文献

一株海洋细菌铁吸收调节蛋白(Fur)基因的克隆和表达

对海洋细菌来说铁是一种必需元素,对铁吸收与利用的调控在细菌生长和防止铁毒性中起重要作用,在革兰氏阴性细菌中,铁的代谢由铁吸收调节蛋白(Fur) 来调控.橙色交替单胞菌是1种具有广泛抑菌谱的有益菌,本研究克隆表达了橙色交替单胞菌的fur基因,并分析了其相关特征.该基因序列中包含447bp的开放阅读框,编码148个氨基酸残基的蛋白,推导的相对分子量为16.637 kD,与GenBank报道的革兰氏阴性菌的相应序列同源性很高,其中氨基酸序列的中段从G47位至D104位,为高度保守区域.在限铁和富铁培养条件下铁载体分泌量的结果分析表明,fur基因可在大肠杆菌中大量表达,为进一步研究Fur蛋白的结构与功能打下基础.  相似文献

为进一步研究溶藻弧菌碱性丝氨酸蛋白酶(ASP)蛋白生物学活性和功能,将构建的pET23d-asp在大肠杆菌BL21(DE3)中表达,对表达产物进行纯化分析,并优化表达条件。结果表明:在咪唑洗脱缓冲液浓度为100 mmol/L时,表达的可溶性ASP被大量洗脱,纯化的ASP相对分子质量约为42 000,具有蛋白活性;在诱导温度28℃、异丙基-β-D硫代半乳糖苷(Isopropyl-β-D-thiogalactopyranoside,IPTG)浓度1 mmol/L及诱导时间16 h时,表达的活性ASP蛋白最多。  相似文献
